چکیده
Molecular techniques are used today in many areas and have increased the confi dence level of studies. Th e aim of the study was to compare the classical morphological characters of coccoid green algae under fi eld and culture conditions by using modern molecular analysis. To achieve this aim, we fi rst isolated the coccoid green algae from Liman and Cernek lakes. Th e morphological variations of green algae were observed under culture conditions and the phylogenetic relationships of these strains were defi ned according to the 18S rRNA gene sequences. According to sequence analysis, 2 of the isolated species had high similarity to Scenedesmus subspicatus (98%) and Desmodesmus sp. (100%).

Xylochloris irregularis gen. et sp. nov. (Trebouxiophyceae, Chlorophyta), a novel subaerial coccoid green alga
The phylogenetic diversity of subaerial coccoid green algae remains still poorly explored. We characterised in detail two unicellular green algae found on tropical trees in Singapore. Light microscopy revealed morphological identity of these two strains. Depending on the age of cultures, the cells were spherical to cylindrical, and ranged in size from 13.5 to 20.5 mm. Each cell contained a pyre...
متن کاملJenufa Gen. Nov.: a New Genus of Coccoid Green Algae (chlorophyceae, Incertae Sedis) Previously Recorded by Environmental Sequencing(1).
The diversity of eukaryotic microorganisms is far from fully described, as indicated by the vast number of unassigned genotypes retrieved by environmental sequencing or metagenomics. We isolated several strains of unicellular green algae from algal biofilms growing on tree bark in a Southeast Asian tropical rainforest and determined them to be relatives of an unidentified lineage of environment...
متن کاملIsolation, morphological and molecular identification of Dunaliella species in Hoz-e Soltan and Aran-va-Bidgol salt Lakes (Iran)
In this study, Dunaliella has been isolated in Jun. 2014 from Hoz-e Soltan and Aran-va-Bidgol salt Lakes (Iran). The isolates have been identified morphologically with the aid of simple light and invert microscope after isolation and purification on their specific medium (Moh202). By using molecular techniques such as polymerase chain reaction (PCR) and amplification of 18S rDNA gene primers, f...
متن کاملLeptochlorella corticola gen. et sp. nov. and Kalinella apyrenoidosa sp. nov.: two novel Chlorella-like green microalgae (Trebouxiophyceae, Chlorophyta) from subaerial habitats.
The diversity of green microalgae in subaerial habitats remains largely unexplored and a number of new genus- and species-level lineages have been discovered recently. The traditional green algal genus, Chlorella, which accommodated coccoid unicellular green algal species with globular to oval cells, reproducing entirely by autospores, has been found to be polyphyletic. In this study, we provid...
متن کاملIsolation and cultivation of endosymbiotic algae from green hydra and phylogenetic analysis of 18S rDNA sequences.
Symbiotic associations are of wide significance in evolution and biodiversity. The green hydra is a typical example of endosymbiosis. In its gastrodermal myoepithelial cells it harbors the individuals of a unicellular green algae. Endosymbiotic algae from green hydra have been successfully isolated and permanently maintained in a stable clean lab culture for the first time. We reconstructed the...
